New developments reveal a molecule with a promising function in terms of cancer treatment

ScienceDaily (Oct. 29, 2012) ? One of the current challenges in terms of cancer treatment is how it can be best adapted to patients: Today the emphasis is on personalised treatment (factoring in genetic and metabolic profiles). In response to this growing need for personalisation, there is an increasing demand for fundamental research to develop adapted future treatments. Researchers from Inserm and CNRS from the Institute for genetics and molecular and cellular biology (IGBMC) and from the Research Institute at the Strasbourg school of biotechnology (Irebs) have focussed their efforts on PARG, currently thought to be a promising new therapeutic target in the treatment of cancer. Their work has revealed the role of this molecule in regulating gene expression.

The results were published on 25 October 2012 in the journal Molecular Cell.

Cells are subjected to various stresses throughout their life. Some of this stress can damage DNA. Fortunately, cells have several mechanisms used to repair these lesions. Breaking two DNA strands is one of the most serious afflictions a chromosome can suffer. The cell must repair this break if it is to continue dividing. Repair actions are either performed in an optimal manner, and the cell resumes its normal division cycle, or the lesion is not repaired correctly, causing the cell to die or the appearance of an anomaly that may trigger cancer.

One of the repair mechanisms used is poly(ADP-ribosyl)ation. In this mechanism, some molecules (PARPs) detect DNA breaks and cause poly(ADP-riboses) synthesis that binds with proteins, thus initialising the lesion repair system. As such, this system can act as a 'saviour' if the repairs are correctly completed, but, it can be equally damaging in the event of incorrect repair.

In the case of cancer, to ensure the cells are destroyed, PARP inhibitors are currently undergoing clinical tests as therapeutic adjuvant used to increase the receptivity of cancer cells to specific chemotherapies.

In terms of fundamental research, researchers know that the poly(ADP-ribosyl)ation mechanism is reversible and is closely regulated by Poly(ADP-ribose) glycohydrolase (PARG). PARP and PARG thus seem to make up a tandem of molecules dedicated to maintaining genome integrity. PARG targeting has proved to potentiate the action of genotoxic agents, making this molecule a promising new therapeutic target in the treatment of some cancers, as is the case for PARP.

However, little research has been conducted into PARG, but in light of its close links with PARP, researchers are now taking a very close look at its functions.

In this new work, the researchers have demonstrated that in addition to its genome repair role, PARG was involved in modulating the cell's transcriptional activity.

Given the keen current interest on PARP and PARG inhibitors in the treatment of cancer, it is essential to gain accurate knowledge of the functions and action modes of these promising therapeutic targets, as well as the consequences of their invalidation. This study is the first to highlight how PARG action mechanism regulates gene expression.

BARCELONA, Spain (AP) -Lionel Messi scored twice to help Spanish leader Barcelona roll to a 5-0 win at Rayo Vallecano on Saturday, as coach Tito Vilanova set the best start for a first-year coach in the 113-year history of the Catalan club.

Cesc Fabregas set up David Villa's 20th-minute opener to set Barcelona on course. Messi scored his 16th and 17th goals in all competitions in a second half that also saw Xavi Hernandez and Fabregas find the net.

Barcelona's eighth win in its first nine games kept it unbeaten and lifted it three points ahead of Atletico Madrid before its game against last-place Osasuna on Sunday.

The no-nonsense win also let Barcelona keep the pressure on Real Madrid by opening up an 11-point gap ahead of the defending champion's tough test at Mallorca.

When Was the First Video Game Tournament?

Atari isn't the only one with a birthday in October. 40 years ago on October 19th, the first video game tournament was held in California at Standford University.?

Video game tournaments are common these days. In fact, competitions in video games is so common that we don?t even have to leave the house to participate anymore. We can just join an online tournament or fight opponents all vying for supremacy on a ladder.

Gamers can just log in to their favorite MMOs, consoles, computer games, or even tablets and cell phones and participate in any number of challenges, competitions, ladders, and all sorts of competitive-minded activities. The competition isn?t just your town, but international. There are even professional gaming associations around now that police proper play.

But it wasn?t always this way. It used to be that video games required you to be present, ready to perform live in front of audiences. With so many people participating in video game tournaments now, when did all of this begin?

Apparently it all began on October 19, 1972 at Stanford University in California.

Walter Day, the now-retired official score keeper of the video game industry and founder of the score-keeping website, Twin Galaxies, recently declared tha

t the very first video game tournament was sponsored by?Rolling Stone?for the video game?Spacewar! on October 19, 1972. This tournament was titled ?The First Intergalactic Spacewar! Olympics" and?the music magazine Rolling Stone?offered participants beer as well as prizes.?

The tournament was played at Stanford's Artificial Intelligence Lab and had two different events: "free-for-all" and "singles." Bruce Baumgart won the ?free-for-all? competition and Slim Tovar secured the singles bracket.

Twin Galaxies?calls themselves the official high score keepers of video gaming. They have been the focus of a number of video game documentaries like?King of Kong?and?Chasing Ghosts: Beyond the Arcade?and have been responsible for a number of video game related entries to the Guinness Book of World Records. To commemorate this event, Twin Galaxies released a new card for their growing set of video game history trading cards.

Stocks higher after strong retail sales report

(AP) ? Stocks are opening higher on Wall Street after a strong report on retail sales.

The Commerce Department says retail sales rose 1.1 percent last month after a 1.2 percent increase in August. Those are the largest gains since October 2010 and could reflect growing confidence in the economy.

Just after the opening bell on Wall Street, the Dow Jones industrial average is up 26 points at 13,355. The Standard & Poor's 500 index is up three at 1,431. And the Nasdaq composite index is up 10 at 3,054.

Citigroup stock is opening higher after the bank beat Wall Street earnings estimates.

My Weapons of Business War, Circa 2012

Todd SchnickThis annual exercise has proven to be quite popular, wherein I review the tools I use to do my work.

Now in my third year (you can find 2010 and 2011 below), I am finding this process helpful in analyzing and determining which tools are most important to me, and the ones that aren?t making the cut.

And in the end, with the ones that are making a difference, I do two things: One, strive to get better at utilizing them, and learn strategies to maximize their impact on my work. Far too often, we only use a small percentage of a tool?s capacity.

You don?t necessarily have to write a post on it, but I?d recommend performing an audit of your professional tools. It does sharpen the saw.

1. WordPress. WP has been my blogging platform of choice since 2008. Easy to use, and great for SEO. All of my companies, and all of my podcast landing pages, use WordPress.

2. Dropbox. As you know, I host and produce dozens of podcasts each week. Dropbox is an amazing tool to store all this audio and ease the transfer amongst my team for editing. In fact, just this weekend, I have moved 90% of my documents and files to Dropbox, as a means to have a backup of all my digital property. And oh so simple to use, and the Dropbox apps for both iPhone and iPad are great, making it VERY easy to view and manage files from those mobile devices.

3. Skype. My preferred vehicle to record my podcasts and video interviews over the internet. Simple, easy-to-use, and syncs very nicely with Call Recorder (seen below). Personally, I use Skype more for client and business conversations than I ever do my iPhone. And the ability to conduct Skype sessions on both my iPad and my iPhone continues to improve.

3.5. Google Hangouts. Just starting to ramp up my usage of Hangouts, and so far, finding great value there. The audio quality of actually quite good, and the ability to enable the Hangout to automatically upload video to your YouTube channel is a simple and easy way to generate and publish content. We may be using Hangouts more and more to record internet-based podcasts.

4. iPhone. A year ago, I was still using a Blackberry and my FlipCam. Man. What a difference. My Flip crashed my Macbook, and I never used it again. And not long after, I bought the iPhone 4S.

5. iPad. I love this thing. In fact, I wish I could run my business on it. But I cannot. So, I consume on this device. I read blogs, scroll thru my LinkedIn connections here, and with my Kindle app, this is mostly where I read my books.

6. Macbook Pro. Will be switching to an Air sometime in 2013. I love this computer, more than any other in my lifetime. Spend most of my time on this writing and editing all my podcasts and radio shows.

7. Moleskine notebooks. If I could only keep one thing on this entire list? It would be my Moleskine notebooks. I just love writing in these. This is where I sketch ideas, list To Dos, makes notes during radio broadcasts. Usually, everything jotted down here ends up strategically placed for action and execution.

8. Uni-ball Vision Fine point pens. I?ve tried a lot of pens over my years. This pen works perfectly on my Moleskines, and is easy to write with. Black ink, btw.

9. Google Drive. Formerly Google Docs, this is where I store spreadsheets, proposals, memoranda, invoices, and much of my writing. The ability to share these docs with others makes for very simple collaboration and co-creation.

10. Gmail. My email platform of choice. Simple. Easy. Syncs well with all of the other stuff I do within Google World, and functions nicely with my mobile devices. I particularly like the archive and search functions behind the service, and continue to learn hacks to make it even more impactful.

11. Blubrry. This is the platform where I store all of my finished and edited podcast audio, and through this service, I stream all of my podcasts to iTunes.

12. Evernote. This is where I store ideas, photos, videos, and business cards. On my browser, I have added the Evernote extension to Chrome, and whenever I find an article that I want to keep, I merely click a button, and the file is saved to Evernote. This is also where I keep digital photos of the business cards I receive and want to keep. I can pull them up on my mobile devices when I need them.

13. Swiss Army SwissGear Airflow Backpack. I have to be pretty mobile going from studio to studio, coffee shop to coffee shop, and especially when broadcasting from trade shows. But I dig this backpack!

14. Notational Velocity. This app on my Macbook is where most early drafts of my writing are started. A great, simple little writing tool. No frills.

15. Google RSS Reader. RSS is getting a little old-school these days, but I still love it. This is where I consume blog posts (Macbook and iPad), where I stream news, monitor LinkedIn connections, and where I monitor personal and client Google Alerts. For me, almost indispensable.

16. 37Signals? Highrise. This is my personal CRM system. Tons of various CRM products on the market. Tons of products that are too complex, rigid, unncessary for my purpose. This is a simple, easy-to-use project that works well on both my Macbook and by iPhone.

17. Call Recorder. This is the app (for Macs) I use to record my conversations (both audio and visual) on Skype. This produces that file that I ultimately edit and use to publish my radio shows.

International Business Organization Development Factors For

International Business Organization

Export/International business can take on many challenges as it unfolds. Goal should be to construct the company's strategic building blocks, using it's assets to support international opportunities. The outline is formatted from a more basic approach, increasing through stages of complexity. There will be points below that overlap and dovetail. Some corporations may have already touched on many of these points. The examples below are geared toward the food/protein industries. Nevertheless, the concepts are transferable to many others. Below are organizational ideas for consideration:

I. International Business Unit Establishment- Create P&L. A. Budget to encompass 3-5 key trade shows and conferences that support the geographic strategy.

- Restaurant Chain Shows (Subway, McDonalds, etc.)- Important shows that demonstrate a company's willingness to globally expand with the chains.

- Distributor Shows.

- Trade organization conferences. Provide key insights to new emerging markets and trends i.e., USMEF, USDEC, etc.

B. Forecasting- By product category and market to determine business profitability.

C. Expenses- Identify expenses against the business. Be fiscally prudent.

II. Geography- Are the most immediate markets being efficiently addressed by export? Begin with the immediate opportunities i.e., target nearest or import friendly international geographic markets.

A. Canada-

B. Mexico-

C. Caribbean-

D. Domestic Exporters-

III. Export Product Portfolio- Product's export potential? What are the popular US items sold? Using meat products as an example:

A. Pork- More than likely highest export potential.

B. Poultry- Certain drawbacks (Avian viruses), but often has the necessary price points for market entry.

C. Beef- Still questionable into many overseas markets (BSE). Slowly improving.

D. Other- Veal and lamb offer the specialty items often sought in many of the smaller boutique markets i.e., Caribbean. Should be a high margin opportunity?

IV. Utilize and maximize current customer base. Grow internationally with domestic customers.

A. Chains- What chains are currently being serviced (i.e., McDonalds)? What are the int'l springboard applications of those chains?

B. Distributors- GFS,US :: GFS, Canada; Sysco, US :: Sysco, Canada...Sysco, Export C. Schools- Offer products supplied to the US to Puerto Rico. Puerto Rico, Virgin Islands and Guam have same requirements.

D. Retail.

V. Expand Geography- Be first in emerging markets. Chains, trade organizations and trade shows will assist in breaking into new venues.

A. Australia, open to US pork imports. US plants must be Australian approved.

B. Brazil and Argentina- as economies improve, so should pork imports.

C. Middle East- Israel.

D. Asia- SE Asia, Latin America.

VI. Product adaptation-

A. A commitment to international product customization. Overcome import non-tariff barriers through product modification.

B. As point "A" is evaluated, determine volumes and pricing with the customer completing the value proposition.

C. New protein introduction- Growing US Hispanic community looking to satisfy traditional diets i.e., goat. US ranchers begin to emerge from their traditional ranching habits to fill a consumer need. Shift creates new export opportunities.

D. Profit Margin/Revenue Growth- Theoretically, there is no competition for custom production and margins should reflect business value.

VII. Resource utilization-

A. R&D efforts to meet a qualified opportunity. Example, a 51% breaded product can be exported to Canada vs. a product with less than 50% breading.

B. Account Managers- Joint calls on corporate to further support the chains international expansions.

Distributor Managers- Joint calls in evaluating immediate opportunities extending across borders. C. School Managers- Joint calls in US territories to expand and maximize product presence.

VIII. International Partnership Arrangements. Partnering/Joint Ventures with like businesses overseas. Some ideal targets are Japan, Australia, Mexico, China. Key defining terms...product novelty, business profitability, uniqueness, pricing, product demand, market distribution, language understanding, product understanding. If there is a commitment from an overseas manufacturer who understands the product/species, but lacks certain manufacturing capabilities, a partnership should be suggested.

A. Responsibility considerations :

o Raw Material Hedge

o Currency Hedge

o Brokerage Agreement

o Time lines

o Production Capacity

o Legal contract/Export Insurance

B. Partner's Responsibilities:

o Volume Projections

o Co-Pack Agreement

o QA Plant Approval

o Currency Hedge

o Other product opportunities

Notice currency hedge may fall under both and is open for negotiation. It depends on relationship's strength. Many times it should be for the account of the partner. An exception may be made to consummate the deal, or as a long term service insuring a yearly contract renewal.

IX. Licensing- Often used as a barometer in evaluating potential opportunities and minimizing immediate risks.

A. Brand Licensing- What is the true value of a certain brand in an international market? Would be determined by the partner company in that country.

Example. What was the value of the Parkay brand in Canada? Became the second best Canadian margarine brand. Produced by Parmalat in Canada. Brand was licensed by ConAgra US.

B. Technology- Minimizes capital overseas investment, while transferring US production technology.

X. Mergers and Acquisitions- Up to this point a corporation may be supplying and evaluating their export potential. Simultaneously, it should be considering the business worthiness of certain key markets. Ultimately, it may consider investment in those markets.

A. Partnership/JV company may be ripe for buy-out.

B. Margin potential internationally warrants an acquisition for corporate diversification purposes. C. Many similarities i.e., language, business culture, profitability, increased product demand from growing middle class, business supporting political environment.

D. Overcome stringent food import barriers i.e., EEC. Example- Companies have improved international exposure, opening manufacturing plants within the EEC. An example has been the recent purchase of Sara Lee European brands by Smithfield.

XI. Summary- These idea compilations are based on 20 years of international business experiences with four major corporations and an MA in International Business. No one size fits all. The outline can be used to build new profitable opportunities that may not otherwise have been realized or fully exploited.

(AP) ? Republicans Mitt Romney and Paul Ryan went back to school on Saturday to rally college students in all corners of all-important Ohio and hammer at President Barack Obama for going easy on China over unfair trade practices. Obama took precious time off the campaign trail to practice for the next debate against his GOP rival.

It was an unspoken acknowledgment of the importance that Obama attaches to upping his game in Debate No.2 that the president is largely dropping out of sight for five straight days in the final weeks of the race to prepare for Tuesday's encounter in Hempstead, N .Y.

Even while cloistered for debate prep at a sprawling resort in Williamsburg, Va., though, the president didn't completely cede the spotlight to Romney. His weekly radio and Internet address highlighted the Obama administration's work to revive the U.S. auto industry ? a message aimed squarely at working-class voters in manufacturing-heavy states like Ohio.

Romney, for his part, told a crowd of more than 3,000 people at Shawnee State University in Portsmouth that Obama was ducking an important decision on whether China is manipulating its currency to gain a trade advantage. A decision was due on Monday, but the Treasury Department said Friday the decision won't come until after global finance officials meet in early November. That means a decision is unlikely before the Nov. 6 election.

"It's time for us to stand up to China for their cheating," Romney declared. "It's got to stop."

Romney framed the issue squarely as a matter of jobs, saying cheap Chinese products were driving American companies out of business.

"We've got to get those jobs back and make trade to be fair," Romney declared.

Ryan, too, criticized the administration for failing to hold China accountable for its trade practices. During a morning appearance in northeastern Ohio at Youngstown State University, he told a crowd of about 1,400 that his hometown of Janesville, Wis., was much like theirs ? a "blue-collar, factory town" where the struggles of the auto industry hit home hard.

Ryan said the president had led the country toward a higher national debt, steeper taxes and insufficient job growth.

"We can't keep going down this path," he said. "We can't keep accepting this is the new normal."

The Wisconsin congressman then hopscotched to Bowling Green State University, in the northwestern part of the state, where he grabbed a bratwurst with mustard at the college Republicans' tailgate party before the school's football team took on his alma mater, Miami (Ohio) University.

The Obama campaign dismissed the Republicans' tough talk on China as nothing more than talk.

"Mitt Romney will never crack down on China's cheating ? just look at his record," Obama campaign spokesman Danny Kanner said in a statement. He said Romney had opposed Obama administration efforts to impose tariffs on Chinese-made tires and had invested in companies that shipped American jobs to China.

Obama's campaign upped its celebrity quotient as the two sides claw for any advantage in a tight race: Actor Morgan Freeman's commanding voice narrates a new ad telling voters that Obama has met the nation's challenges and "the last thing we should do is turn back now."

On Thursday, Bruce Springsteen will team up with former President Bill Clinton to rally Obama voters in Parma, Ohio, in what will be the singer's first political appearance this campaign. "The Boss" plans a second event Thursday, in Ames, Iowa. Springsteen campaigned for Obama in 2008, too.

Both sides are devoting huge time and effort to Ohio, this year's battleground to end all battlegrounds, where polls show Obama with a slight edge over Romney. Saturday's emphasis by the two sides on the auto industry and manufacturing jobs was designed to connect with blue-collar voters there.

Both sides also are keenly aware of the importance of this year's series of three presidential debates. Romney's strong performance in the first debate on Oct. 3 gave his campaign a much-needed jolt of energy, and the GOP nominee said his campaign still had post-debate momentum over Obama.

"His campaign is about smaller and smaller things, and our campaign is about bigger and bigger crowds, fighting for a bright future," Romney said on warm, sunny fall day.

Joe Biden's aggressive counterpunch in a debate with Ryan on Thursday cheered Democrats, but some critics thought the vice president overdid the theatrics with his frequent eye rolls, headshakes and broad grins suggesting incredulity.

Obama's campaign has acknowledged he didn't practice enough before his widely panned performance in the first debate. Some sessions were cut short, others canceled altogether, mainly because of developments in Libya, where four Americans were killed at a U.S. consulate.

The campaign has resisted calls from some Democrats to shake up Obama's debate team. Senior advisers David Axelrod and David Plouffe, along with former White House officials Anita Dunn and Ron Klain, still are running the preparations.

The president may have picked up a few pointers from Biden's debate with Ryan. Obama watched the vice presidential debate from aboard Air Force One and would chime in when Biden made a strong point.

"That's a good one," Obama said, according to aides.

Tuesday's town hall-style debate at Hofstra University will have an audience of about 80 undecided voters selected by the Gallup Organization. Moderator Candy Crowley of CNN will select from among questions on foreign and domestic policy submitted by the audience. The final debate, covering foreign policy, will be Oct. 22 in Boca Raton, Fla.

Romney spent nearly four hours Saturday morning at a Columbus hotel preparing for the next debate, then boarded his campaign bus for Shawnee State. From there, the Romney bus headed for Lebanon in southwest Ohio.

The Obama campaign isn't leaving Ohio unguarded for long: Michelle Obama will visit Delaware and Cleveland on Monday and the president will be in Athens on Wednesday.

To help keep Democratic ads like Freeman's new appeal for Obama up and running in the hotly contested battleground states, the president dispatched Biden to a pair of private fundraisers in Connecticut and New York on Saturday.

Dr. Susan Curtis, professor of history at Purdue, will present ?The Politics of Memory: Archives, Historic Preservation and Lessons of the Past? at 7:00pm, Thursday, October 18th in the Morgan University Center Ballroom B and C.

The event is sponsored by the Woodward Library Society, the Wilbur N. Daniel African American Cultural Center and the APSU Honors Program. There is no cost, and it is open to the general public. Refreshments will be served at the conclusion of the presentation.

In addition to her teaching duties, Curtis is director of American studies at Purdue University. Her recent research has focused on the meaning of American culture in a multiracial and multiethnic society.

Curtis earned a B.A. at Graceland College and an M.A. and Ph.D. at the University of Missouri. She teaches courses in U.S. cultural history, American studies, and the history of religion in America.

Curtis has published three major works: ?A Consuming Faith: The Social Gospel and Modern American Culture? (1991), which demonstrated the interplay between sacred and secular realms in the reformulation of Protestant thought and practice between the 1880s and 1920s; ?Dancing to a Black Man?s Tune: A Life of Scott Joplin? (1994), which used the life of Scott Joplin to examine the cross-racial collaboration at the turn of the century that resulted in ragtime, a quintessentially American popular music; and ?The First Black Actors on the Great White Way? (1998), which places the landmark event noted in the title in the cultural context of the U.S. on the eve of World War I.

The Woodward Library Society is an organization of library friends dedicated to the advancement of APSU?s Felix G. Woodward Library. The purpose of the society is to strengthen the resources of the Woodward Library and to increase awareness of those resources.

Founded in 2009, the society is a nonprofit organization affiliated with the APSU Foundation. Membership is open, upon payment of annual dues, to any person, business, or organization that shares in the purpose of the society.

Satellite broadband gets millions more Africans online

The launch of a new satellite will provide broadband access for millions in Africa, without having to rely on fault-prone undersea cables

LIKE many cybercafes in the centre of the Ugandan capital, Kampala, the Amazon Digital Internet Caf? is always jam-packed. Run by Annette Byaruhanga and her husband Ernest, the venue can make up to 400,000 Ugandan shillings ($155) on a good day.

But every now and then there is a major problem. When the country's undersea communications cables are damaged - and they often are - it causes havoc for local people and businesses. During a recent hiccup, Annette Byaruhanga lost business for two weeks. "We were severely affected by disruption of broadband service when the undersea cable was ruptured," she says. "The damage spoiled our reputation. Our customers lost confidence in our caf?."

There could soon be a solution. A recent satellite launch promises to revolutionise broadband in East Africa, helping to bring high-speed internet to some of the millions in the region still without it.

Many in the region, like Byaruhanga and her husband, feel let down by the undersea fibre-optic system that connects some African countries to Europe and Asia. The multilayered, armoured cables have been cut on several occasions by shipping, and there are repeated outages. The most recent serious incident was in February this year and was due to a ship's anchor. It left Ugandans without internet access for days.

For both eastern and southern Africa, things are about to change. The small HYLAS 2 satellite, developed by Avanti Communications, based in London, was launched in early August from the European spaceport in Kourou, French Guiana, South America. The satellite completed its orbital testing in early September and is almost ready to begin commercial services.

Once HYLAS 2 is transmitting, individuals and small businesses with personal satellite dishes will be able to receive data at up to 20 megabits per second. Telecoms companies and big multinationals will get faster speeds, says Avanti chief executive David Williams.

The socio-economic benefits could be enormous. A report issued last month by the Commonwealth Telecommunications Organisation says that a 10 per cent increase in broadband availability generates economic growth of 1.38 per cent, nearly twice the impact of the same increase in availability of fixed-line telephones. It also says that, as of June this year, some 341 million people in sub-Saharan Africa would have had to travel at least 50 kilometres from their homes to reach a place with broadband internet. And it adds that an estimated 40 per cent of sub-Saharan Africa will not have access to broadband services unless there is greater use of satellites.

Satellite broadband, often heralded as the future of internet access in the west, could soon be booming in Africa. The launch of a second HYLAS satellite is now scheduled for 2015. Together, the two satellites will bring up to 100 gigabits of capacity to Africa, claims Williams. While it is almost certain to cost more than existing broadband access, its greater reliability will make it a must-have for many of Uganda's small businesses.

"With reliable satellite broadband, even if it costs more, I would be sure that the internet link is up and running all the time," says Byaruhanga.

SANAA, Yemen (AP) ? A drive-by shooting Thursday that killed a top Yemeni security official who worked at the U.S. Embassy in Sanaa has raised concern that al-Qaida militants here are bouncing back and getting bolder after suffering defeats this year in a U.S.-Yemeni military offensive.

Al-Qaida in Yemen has carried out a string of assassinations of top security and military officials and deadly suicide bombings in recent months. Security officials said they believe it has a hit list to kill more in an attempt to paralyze Yemen's new leadership installed this year and throw the anti-al-Qaida assaults into turmoil.

The new killing also raises the possibility the group could turn its assassination campaign against American interests as well. The group has sought to ride the recent wave of anger against the U.S. over an anti-Islam film by calling for attacks on American and other foreign diplomatic missions.

In Thursday's attack, embassy security official Qassem Aqlani was on his way to work when a masked gunman on a motorcycle opened fire on him and fled the scene, officials said. The attack was near Aqlani's home in western Sanaa, while the embassy is located in the eastern part of the capital. Aqlani, who was in his 50s, had been working for the embassy for 11 years, the U.S. State Department said.

Yemeni security officials said the attack was similar to previous assassinations by al-Qaida, although they said it was too early to confirm the group's involvement. The officials spoke on condition of anonymity because they were not authorized to talk to the press.

There are increased concerns over security at American embassies in the region after last month's deadly attack by militants against a U.S. diplomatic mission in Benghazi that killed the U.S ambassador to Libya and three other Americans. Protesters also stormed several U.S. embassies in Arab nations ? including the one in Sanaa ? in outrage over the film denigrating the Prophet Muhammad.

An elite team of around 50 U.S. Marines sent to bolster security at the Sanaa embassy after the Sept. 13 attack was scheduled to leave the country later Thursday, Yemeni officials said. It was not clear whether Aqlani's slaying would affect those plans.

There was no sign of increased security around the U.S. Embassy in Sanaa, which has already been heavily fortified with new protective measures amid the turmoil surrounding last year's uprising against then-president Ali Abdullah Saleh. Security was tightened even more after the protesters stormed the compound last month. They climbed over the walls, damaged some buildings and took down the U.S flag before they were driven out by Yemeni soldiers.

Since then, the main road that leads to the embassy is sealed off by cement blocks and three checkpoints limit access, with cars scanned for explosives. Yemeni troops and armored vehicles also are stationed near the building.

But officials said they believe it was not the anti-Islam movie that led to Thursday's killing, but that it was part of al-Qaida's campaign of retaliation for heavy blows the group has suffered in recent months.

Washington considers the Yemen-based al-Qaida in the Arabian Peninsula, or AQAP, to be the most dangerous offshoot of the terror network after it was linked to several attempted attacks on U.S. targets, including the foiled Christmas Day 2009 bombing of an airliner over Detroit and explosives-laden parcels intercepted aboard cargo flights last year. The group made major gains in Yemen last year amid the uprising that led to Saleh's ouster. Its militants and their allies seized several towns and cities in the south of the country.

But Saleh's successor Abed Rabbo Mansour Hadi has worked closely with the U.S. in pushing the group back. An offensive by Yemeni troops backed by U.S. airpower and advisers drove militants out of the southern cities and into mountain refuges. Airstrikes, believed to be by U.S. drones, have killed a number of key al-Qaida operatives, including AQAP's deputy leader, Saeed al-Shihri, and six others traveling with him in a car last month.

In retaliation, the group appears to have changed tactics from trying to hold territory to carrying out deadly attacks.

Two weeks ago, a top intelligence official, Col. Abdullah al-Ashwal, was killed in a drive-by shooting in Sanaa by his own informant, who appeared to be working for al-Qaida.

Over the summer, a suicide bomber killed Maj. Gen. Salem al-Quton, the top commander of the southern region who led the war against al-Qaida. Col. Abdullah al-Maouzaei, charged with hunting down members of al-Qaida, was killed when his vehicle blew up as he turned on the ignition outside his home in the southern port city of Aden. A top intelligence official in Sanaa and Omar Barsheed ? who headed the Military Academy and was nominated to head presidential guards ? both died in similar bombings. The press secretary of the president escaped an assassination attempt.

One of the worst attacks took place on May 21 when a suicide bomber ? who turned out to be a son of a colonel? blew himself up in the middle of military rehearsal killing nearly 100 of them. A month later, al-Qaida suicide bomber detonated his explosives among a crowd of Yemeni police cadets, killing at least 10.

"These operations aim at paralyzing the military forces and breaking their morale," said Mansour Hayel, a political analyst.

Officials said that detained al-Qaida members confessed that the slain officials were on a hit list that includes many of Hadi's men, particularly those he depends on in his war against al-Qaida.

Hadi has reached out to tribal leaders in the south, trying to secure their help against al-Qaida, after many of its militants who fled this year's offensive took refuge in tribal-controlled areas in the mountains in the southern and eastern parts of the country. During the offensive, the Yemeni military has often relied on local tribal militias for help.

On Tuesday, while meeting with tribal leaders from Shabwa province ? a one-time stronghold of al-Qaida ? Hadi pressed for unity with the government, warning that the government "will not tolerate anyone who helps al-Qaida."

But al-Qaida is sending signals to the tribes as well. Authorities on Tuesday discovered three decapitated bodies dumped in an open-air market in the eastern province of Marib.

Local media reported that CDs found next to the bodies showed the men confessing to being government informants against al-Qaida and admitting they had put tracking devices on cars that became targets for U.S. drone strikes. One of the men said he worked for a tire repair shop and used to plant chips in militants' vehicles while replacing their tires.

Romney: US was 'attacked successfully' in Libya

ASHEVILLE, N.C. (AP) ? Republican candidate Mitt Romney said Thursday the attack on the U.S. Consulate in Libya is an issue in the presidential campaign in part because Americans wonder why it took the Obama administration so long to acknowledge it was a terrorist act.

Romney was responding to a charge by President Barack Obama's campaign that the Republican ticket has been politicizing the attack, which killed the U.S. ambassador to Libya and three other Americans.

At a rally in North Carolina, Romney read aloud comments from Obama spokeswoman Stephanie Cutter about the attack. "The entire reason that this has become the, you know, political topic it is, is because of Mitt Romney and Paul Ryan," Cutter told CNN.

"No, President Obama," Romney said. "It's an issue because this is the first time in 33 years that a United States ambassador has been assassinated. Mr. President, this is an issue because we were attacked successfully by terrorists on the anniversary of Sept. 11. President Obama, this is an issue because Americans wonder why it was it took so long for you and your administration to admit that this was a terrorist attack."

In response, Obama campaign spokeswoman Lis Smith said Romney was trying to "score cheap political points."

The back-and-forth comes a day after Republicans used a politically charged House hearing to confront State Department officials about security at the U.S. consulate in Libya and assail the Obama administration's early response to the killing of the ambassador and three other Americans there.

U.S. Ambassador Chris Stevens died in what administration officials now describe as an act of terrorism.

In statements immediately after the attack, neither Obama nor Secretary of State Hillary Rodham Clinton mentioned terrorism, though Obama referred to "acts of terror" in his Rose Garden statement the morning after the assault. Both Obama and Clinton gave credence to the notion that the attack was related to protests about the privately made anti-Islam video.

"Some have sought to justify this vicious behavior as a response to inflammatory material posted on the Internet," Clinton said on the night of the attack. "The United States deplores any intentional effort to denigrate the religious beliefs of others. Our commitment to religious tolerance goes back to the very beginning of our nation. But let me be clear: There is never any justification for violent acts of this kind."

The day after the attacks, State Department officials described them in a conference call with reporters. They outlined a prolonged assault that involved attacks on two different buildings at the Benghazi consulate, with Stevens and other officials trying to escape from one building only to be pinned down by gunfire in another.

Five days after the attack, U.N. Ambassador Susan Rice said her best information at the time was that it stemmed from a protest that became violent.

White House spokesman Jay Carney told reporters Wednesday that in hindsight "there is no question that the security was not enough to prevent that tragedy from happening. There were four Americans killed."

Some photos of what may be the next Google Nexus smartphone leaked this earlier?week, but now a whole prototype may have slipped out. Rumor is that this LG-made device will be officially unveiled on Oct. 29.

This latest bit of Nexus-related gossip comes via the folks at?Russian news site Onliner, who say that they got their hands on what appears to be a prototype of the next version of Google's Nexus smartphone. The device appears to match the one we've seen in previously leaked images.

According to Onliner's report, the prototype device is codenamed "E960" and runs Android 4.1.2. It supposedly has a 1.5-GHz quad-core processor, 2?GB of RAM, and 8 GB of storage. Its screen measures 4.7?inches and has a resolution of 768 x 1280 pixels.

It's not clear from the rumor posts to date whether or not the phone is 4G LTE capable, but we'd hope ? and?even expect ??that it does.

Onliner's report reiterates the idea that the next Google Nexus smartphone will be announced on Oct. 29. We would not be surprised if this detail turns out to be true as?that day?would be almost exactly a year from the moment Samsung and Google announced the current Google Nexus.
